Objective

To perform a restriction enzyme digestion of Lambda DNA and visualize the resulting fragments through agarose gel electrophoresis to create a Gel Art pattern.

Materials

Reagents & Samples

• Lambda DNA (New England Biolabs - NEB)
• Restriction enzymes:
  • Lane 1: BamHI, EcoRI, EcoRV, HindIII, KpnI, SacI, SalI
  • Lane 2: EcoRI, KpnI, SalI
  • Lane 3: EcoRV, HindIII
• 10X Restriction Enzyme Buffer (compatible with selected enzymes)
• Nuclease-free water
• DNA Ladder (1 kb Plus, NEB)
• Loading dye (6X)
• Agarose (1%)
• TAE Buffer (1X)
• Ethidium bromide (EtBr) or SYBR Safe (for staining)

Equipment

• Micropipettes (1-10 µL, 20-200 µL)
• Gel electrophoresis apparatus
• Power supply
• UV transilluminator or blue light viewer
• Microcentrifuge tubes (1.5 mL)
